Design: The first step in the generation of a SMRTbell library was fragmentation of genomic DNA to appropriate sizes. Then DNA fragments were damage repaired, end-repaired, and A-tailed. The SMRTbell library was produced by ligating universal hairpin adapters onto double-stranded DNA fragments. After the exonuclease and AMPure PB beads purification steps, sequencing primer was annealed to the SMRTbell templates, followed by binding of the sequence polymerase to the annealed templates. The library was checked with Qubit for quantification and bioanalyzer for size distribution detection. Quantified libraries will be pooled and sequenced on PacBio Sequel II/IIe systems, according to effective library concentration and data amount required.
